If you want to look at this from a "canon" standpoint:
Isabelle always struck me as a net sub. She's extremely subservient, even to her own detriment seeing as she barely sleeps, she's straight up afraid of having to take charge when it comes to ordering people around (when you ask her to confront a villager that's misbehaving), dislikes stepping out of line, and likes when everything is ordered as it should be, which I interpret would extend to how she sees herself, a woman, in a relationship.
Ankha... it would be easy to base her on the general fandom perception, but that's all complete headcanon/fanon. Unlike Isabelle, she's a villager with a generic (snooty) personality. Villager personalities are in part down to how players inject flavor into them, which is how we got our colorful demeaning queen, but even at base, snooty villagers always have that air of superiority about them. They're prone to boss others around.
Either way, Isabelle is way to subservient to be a dom... but dom/sub is not the same as top/bottom, is it? I can definitely picture Ankha as a pillow princess, taking the role as the bottom while using Isabelle's compliant personality to completely boss her around to do all the work, which she would be more than eager to do without ever falling out of line because obeying is kind of her whole thing.
